The Temperamental Blueprint of Life: The Mizāj–Tarkīb–Af'āl Triad in Unani Medicine and Its Convergence with Systems Biology and Epigenetics
Abstract
This study explores the classical Unani triad of Mizāj (temperament), Tarkīb (structure), and Af‘āl (function) as a coherent framework for understanding individuality, health, and disease. Through an integrative, qualitative review, classical doctrines from Avicenna, Galen, and Jorjani are examined alongside contemporary insights from systems biology and epigenetics. The paper interprets Mizāj as the internal qualitative blueprint that shapes structural form (Tarkīb) and functional behavior (Af‘āl), establishing a dynamic link between elemental balance and biological expression. The concept of Harārat Gharīziyya (innate heat) is correlated with mitochondrial bioenergetics and cellular metabolism, while Sittah Ḍarūriyyah (six essential requisites) is analysed in the context of environmental epigenetic regulation. The analysis suggests strong conceptual convergence between Unani principles and modern scientific models of bio-individuality, gene–environment interaction, and personalized medicine. The Unani approach parallels contemporary systems-level understanding of health now emerging in integrative and preventive medicine. The paper concludes that the Mizāj–Tarkīb–Af‘āl triad offers a robust philosophical and translational model, providing new directions for research that links traditional qualitative frameworks with measurable molecular mechanisms.
