Advanced Theoretical Framework for Systemic Senescence Reversal
Abstract
Radical lifespan extension requires overcoming the physiological barriers of delivery, immunity, and oncogenesis. Previous protocols proposed broad immunosuppression and CD4 7 -cloaking, which carry risks of latent viral reactivation and autoimmunity. This paper presents a refined, safety-first engineering framework. We propose the use of miRNA-detargeted AAVs to enforce strict tissue specificity, coupled with an Inducible Caspase-9 (iCasp9 ) Kill Switch to mitigate the risks of CD4 7 overexpression. Furthermore, the protocol incorporates a Prophylactic Antiviral Regimen during aseptic quarantine to prevent opportunistic reactivation of latent pathogens (CMV/EBV). Stochastic modeling updated with these safety variables suggests a viable path to significant healthspan extension with managed risks.
