Abstract

Supplementary Figure S1. Antibody validation by comparison of antibodies. The panels show immunostaining results obtained by two independent MRE11 antibodies. Using HMV328, the stainings showed a strong nuclear MRE11 positivity in all cells of the thymus but an even stronger positivity was discernible in a distinct subset of scattered cells (A), a generally weak nuclear staining in the cerebellum but a stronger intensity in Purkinje cells (B), a decreasing staining intensity with maturation of cells of the spermiogenesis in the testis (C), a decreasing staining intensity from the basal to the superficial cell layers of esophageal squamous epithelium (D), weak or absent staining in syncytiotrophoblastics of the mature placenta (E) and in hepatocytes (F), a distinct nuclear staining in the stomach (G), and a strong nuclear staining of adrenocortical cells (H). Using MRE11 #4895, a comparable nuclear staining was seen in thymus (a), cerebellum (b), testis (c), squamous epithelium of esophagus (d), placenta (e), liver (f), stomach (g), and the adrenal gland (h). An additional excessive cytoplasmic staining of hepatocytes (f) and to a lesser extent in adrenocortical cells (h) was only seen by the antibody #4895 and thus considered an antibody specific cross-reactivity of the antibody #4895. As the cytoplasmic staining of gastric parietal cells by HMV328 was not confirmed by #4895, this staining was considered an antibody specific cross-reactivity of the antibody HMV328. The images A-H and a-h are from consecutive tissue sections. Supplementary Figure S2: Comparison of study data with previous reports on MRE11 IHC. An X “indicates the fraction of MRE11 positive cancers in the present study and dots indicate the reported frequencies from the literature for comparison. Green dots mark studies with >20 positive tumors and yellow dot mark one study with
