A Comprehensive Review on Bioanalytical Method Development: Extraction Techniques, Instrumentation, and Validation Frameworks
Abstract
Bioanalysis is really important in developing new drugs, especially when studying how the drug moves through the body, how it works, how safe it is, and how well it's absorbed. Creating and testing bioanalytical methods helps make sure we can accurately measure the drug and its breakdown products in things like blood, urine, or saliva. This review covers the basics, what’s needed, and the rules to follow when developing and testing these methods. Key steps in the process include checking the drug’s physical and chemical properties, choosing the right equipment for separating substances, improving ways to extract the drug from samples, picking a good internal standard, and setting up sample preparation. Common sample preparation methods like liquid–liquid extraction, solid-phase extraction, protein precipitation, and combined techniques are covered, as well as newer microextraction methods such as SPME, SBSE, and MEPS. The review also looks at modern lab equipment, focusing on LC–MS/MS and UPLC–MS/MS because they offer better sensitivity, accuracy, and work well for analysing many samples quickly. It goes over important validation factors like how well the method works over a range, how accurate and repeatable it is, how specific it is, how much of the sample is recovered, how stable the results are, and how reliable the method is. It also covers different types of validation—full, partial, and cross-validation—and explains how they fit with rules and everyday drug testing. The text also talks about keeping good records, checking that the equipment is working properly, making calibration curves, and following quality control steps to make sure results are consistent and can be repeated. In short, reliable bioanalytical methods are key to keeping data accurate, following rules, and helping with drug research and development. This review covers the main approaches, basic principles, and what regulations expect for good bioanalytical work in today's pharmaceutical field.
