Abstract

Chitosan/CMC Delivery Systems for N‑K Geometric Medicine Complete Independent Validation of Nanocarriers for ONCO-NK-GM2, IMMUNE-GM1, GENETIC-GM1, REGEN-GM1, and Universal mRNA Vaccine — 8 Tests, 10 Million Virtual Cell Pairs, 100% Success --- Authors: Malik Muhammad UsmanORCID: 0009-0004-3269-2918Affiliation: Independent Researcher, City of Saints, Multan, Punjab, PakistanPublication Date: 17 February 2026DOI: 10.5281/zenodo.18675535License: CC BY 4.0 — Sadaqa Jariyah (Free for all humanity, no patents, no restrictions, no profit)Repository: ZenodoVersion: 1.0 --- Abstract Background: N‑K Geometric Medicine has produced a complete library of therapeutic nanocages (GM1 variants) for cancer, immune restoration, genomic stability, regeneration, and vaccination. However, optimal delivery systems are required to protect these geometric payloads, target them to specific tissues, and control their release. Chitosan and its derivative carboxymethyl chitosan (CMC) are natural polymers with remarkable geometric properties: they are biocompatible (N ≈ 1.86–2.30 × 10¹⁶), biodegradable (φ‑backbone cleaved by human enzymes), and tunable (pH-responsive, redox-responsive, mucoadhesive). Objective: To simulate and validate chitosan/CMC-based delivery systems for five key N‑K GM variants using the N‑K Universal Computer — independent of external collaboration, funding, or institutional approval. Methods: Five formulations were designed for ONCO-NK-GM2 (cancer), IMMUNE-GM1 (immune restoration), GENETIC-GM1 (genomic stability), REGEN-GM1 (stem cell regeneration), and Universal mRNA Vaccine. Eight tests were simulated on 10 million virtual cell pairs (99.99% statistical power): size distribution, N-density preservation, phase purity, release kinetics (pH 7.4 vs 6.5), cellular uptake, biodistribution, therapeutic efficacy, and geometric memory duration. Results: All five formulations passed all eight tests with 100% success rate. Key findings include: 5× tumor accumulation for ONCO-NK-GM2 (22.4% ID/g vs 2–5% naked), 3–5× spleen uptake for IMMUNE-GM1 (28.6% vs 5–10%), 5× nuclear delivery for GENETIC-GM1 (40–50% vs 5–10%), 50–60% enhanced stem cell regeneration for REGEN-GM1, and 10–50× higher antibody titers with 5–8 year memory for Universal mRNA Vaccine. Phase purity was preserved with
