Abstract

Withania somnifera (Ashwagandha) is a prominent medicinal plant extensively used in traditional systems of medicine, particularly Ayurveda, for its wide range of therapeutic applications. Recent scientific interest has focused on systematically exploring its phytochemical composition, pharmacological properties, and computational potential to validate traditional claims and support modern drug discovery efforts. Ashwagandha is rich in bioactive constituents such as withanolides, alkaloids, flavonoids, and steroidal lactones, which contribute to its diverse biological activities. Pharmacological investigations have demonstrated its adaptogenic, anti-inflammatory, antioxidant, immunomodulatory, neuroprotective, anticancer, antidiabetic, and cardioprotective effects through multiple molecular pathways. In parallel, advances in computational approaches—including molecular docking, pharmacophore modeling, network pharmacology, and molecular dynamics simulations—have provided insights into the interactions between Ashwagandha derived phytochemicals and key biological targets involved in stress regulation, inflammation, neurodegeneration, and metabolic disorders. These in silico studies complement experimental findings by predicting bioavailability, target specificity, and safety profiles, thereby accelerating lead identification and optimization. Overall, the integration of phytochemical analysis, pharmacological validation, and computational modeling highlights the therapeutic potential of Withania somnifera as a valuable source of bioactive compounds and underscores its relevance in evidence-based herbal medicine and modern pharmaceutical research.
