Simultaneous Estimation of Metformin and Glimepiride By RP-HPLC
Abstract
The simultaneous estimation in RP-HPLC has become standard practice to determine Metformin and Glimepiride together to conduct quality control of combined antidiabetic formulations. Traditional RP-HPLC techniques employ organic solvents like methanol and Acetonitrile which are expensive and dangerous to work with for the user and the environment. The purpose of the study is to explore an eco-friendly, low-cost hydrotropic Sodium benzoate assisted RP-HPLC method for the simultaneous determination of Metformin and Glimepiride. To determine the ideal concentration for peak solubility and ideal retention and chromatographic behavior, we tested several concentrations of Sodium benzoate from 0.1% to 0.9%. Lower concentrations (0.1-%0.4%) were observed to yield peaks with low strength and lower to mid-resolution. Higher concentrations (0.6% to 0.9%) were observed to lead to high viscosity which led to retention times being prolonged). The ideal mobile phase is 0.5% sodium benzoate to Acetonitrile 60:40 which results in sharply peaked chromatograms, ideal symmetrical shapes, and good compartmental separations of the 2 subjects of analytical interest of our RP-HPLC run experiments. The method resulted in lower use than usual for usage of organic solvents and resulted in better performance overall. Thus, our method was able to meet and deliver on the goals of greenchemistry. Thus, the hydrotropic RP-HPLC method is a better and more efficient way to determine the concentrations of Metformin and Glimepiride than current methods and is more reliable, and is more affordable, and is more sustainable.
