Abstract

This collection comprises nine synthesis papers developing and extending the gestational oxygen-timing hypothesis — a framework proposing that disruptions to fetal oxygen supply during specific developmental windows produce latent, multi-system injury through HIF-1α-mediated mechanisms, with particular focus on oligodendrocyte precursor cell (OPC) maturation arrest and consequent myelination-dependent sensory and autonomic phenotypes. The papers span the following domains: Core mechanism: Gestational oxygen-timing alignment and the HIF-1α/OPC maturation arrest pathway as a unifying model for neurodevelopmental conditions including autism. Multi-organ programming: Multi-system gestational oxygen-timing model linking placental insufficiency to staggered organ-specific outcomes across the lifespan. Sensory integration: Insular cortex timing alignment and hyper/hypo sensory oscillation as downstream consequences of partial myelination. Maternal-placental biology: Anti-angiogenic cascade triggers linking maternal vascular dysfunction to fetal oxygen disruption. SIDS and autonomic programming: HIF-1α-dependent autonomic (sympathetic, serotonergic, adrenomedullary) programming and its connection to sudden infant death syndrome. Epigenetic biomarkers: Methylation signatures of gestational hypoxia as potential diagnostic markers. Cancer predisposition: Gestational hypoxia as a mechanism for altered cancer risk trajectories. LMIC natural experiments: Low- and middle-income country birth conditions as natural experiments testing oxygen-timing predictions. Treatment convergence: Therapeutic approaches targeting the oxygen/OPC axis. All papers are evidence-mapped syntheses drawing on published literature. They present testable predictions and proposed study designs. This work is independent and has not undergone formal peer review.
