Abstract

This technical bulletin describes a pooled, quantitative assay for measuring ligand-inducible transcription factor function using the GROQ-seq platform, along with the resulting datasets. The assay links TF–DNA binding to cellular fitness via antibiotic resistance, enabling high-throughput functional profiling through sequencing. It includes datasets for LacI, RamR, and VanR across multiple library types, supporting the development of predictive models for mutation effects.
