Abstract

This technical bulletin presents a pooled, quantitative assay for measuring T7 RNA Polymerase (T7 RNAP) function using the GROQ-seq platform. The assay links enzyme function to cellular fitness via antibiotic resistance, enabling high-throughput functional profiling by sequencing. It reports quantitative measurements across an ~5-fold dynamic range for a dataset of 34,000 variants, including over 12,000 single and 21,000 multi-residue mutants. This work and resulting dataset support the development of predictive models for assessing mutational effects.
