Abstract

This technical bulletin presents a pooled assay for measuring tobacco etch virus (TEV) protease function using the GROQ-seq platform and its first generated dataset. The assay links enzyme function to cellular fitness via antibiotic resistance, enabling high-throughput functional profiling by sequencing. The resulting GROQ-seq TEV Protease assay produces calibrated function measurements spanning an approximate 100-fold dynamic range. Its first generated dataset includes over 18,000 TEV protease variants across 24 different experimental conditions. This work and resulting dataset support the development of predictive models for assessing mutational effects.
