Abstract

This study engaged high performance liquid chromatography-high-resolution mass spectrometry with quadrupole time-of-flight (HPLC-HRMS/MS, QToF) to establish genotypic variation in the comprehensive phenolic profiling of two white grape varieties, Thompson Seedless and Manik Chaman. This investigation identified fourteen distinct phenolic compounds across both cultivars. Characterized each compound based on precursor ion, quality fragment ion, mass error, retention time, accurate mass, and fragmentation patterns. Dihydrocoumarin emerged as the primary marker compound in Thompson Seedless, while Ethyl 2-aminobenzoate in Manik Chaman. Additionally, both varieties identified Arginine, Hydroxycinnamic acid, and Quercetin as major contributors. Phenolic compounds are well known for their antioxidant and anti-inflammatory properties. These compounds play a significant role in cancer prevention and improving health in cardiovascular diseases. This study emphasizes the capability of HPLC-HRMS/MS (QToF) for comprehensive metabolite profiling in different grape varieties. This approach helps to understand the phenolic composition of these two varieties and helps distinguish their individuality from other grape varieties.
