Abstract

Abstract Cryopreservation of sheep semen is an important tool for improving reproduction, but in breeds like Pelibuey and Blackbelly, it remains a challenge because semen loses quality when frozen and thawed. This situation limits the use of genetic material in the Pastaza Province, where these breeds are common and more efficient methods are needed. Therefore, this study sought to evaluate whether the cholesterol-cyclodextrin complex (CLC) could help protect spermatozoa and improve their quality after freezing. The main objective was to determine if different doses of CLC offered better protection of the sperm membrane. A total of 24 straws, which served as the experimental units, were obtained from four treatments: one without CLC and three with 0.5, 1.0, and 1.5 mg, with three replicates. The straws were chilled and then frozen, and macroscopic and microscopic characteristics were evaluated. The results showed that the CLC doses (0.5 and 1.0 mg) helped to better maintain sperm structure after thawing. In conclusion, the use of the CLC complex may be a useful option for improving the quality of frozen sheep semen in Pastaza, and further investigation of other concentrations and with more animals is recommended to confirm these results.
