Current Debate Regarding Laboratory and Natural Origin Hypotheses of SARS-CoV-2: A Critically Balanced Review
Abstract
The emergence of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), leading to the COVID-19 syndrome, triggered a global pandemic with profound medical, social, and economically devastating consequences. The origin of SARS-CoV-2 yet remains one of the most critical scientific questions of our ongoing century. SARS-CoV-2 can be considered a genetically modified organism from the point of origin due to either a zoonotic spillover and/or a laboratory incident. Despite numerous scientific studies, no definitive evidence has resolved whether the virus emerged via a zoonotic spillover and/or started through viral research-related lab-leak modifications. This review follows a structured literature-search approach, integrates mutually supporting and opposing evidence for both hypotheses, outlines limitations, and aims to provide an objective synthesis for clinicians, molecular scientists, and regulatory experts. Current evidence does not conclusively support either a natural zoonotic spillover or a research-related laboratory incident. A cautious, evidence-based approach is essential to support global preparedness for future emerging pathogens.
