Abstract

This chapter proposes the use of three-dimensional (3D) cell culture strategies that, by emulating the physiological characteristics of tissues composed of mammalian cells, allow for the large-scale production of spheroids from human prostate adenocarcinoma tumor cells. It is worth noting that the extended use of 3D cultures has also enabled the investigation of biomolecular mechanisms in cell lines from other types of tumors that are resistant to the conventional treatments, as they can more accurately represent tumor physiology (tumoroids) in an in vitro setting. The 3D culture has proven effective in investigating biomolecular mechanisms of highly aggressive tumor cell lines. Preliminary results indicate that key signaling pathways involved in the maintenance of clonogenesis can be successfully explored using three-dimensional cultures.
