Abstract

A) Percentage of SARS-CoV-2 N positive cells in CF iPSC-airways treated with and without ETI by immunoimaging, expressed as a percentage of total DAPI+ positive cells. Statistical significance was determined as p-value of < 0.05 using Student’s t test (*: p ≤ 0.05; **: p ≤ 0.01; ***: p ≤ 0.001 B) Percentage of SARS-CoV-2 N positive cells in CFTR-Corrected iPSC-airways treated with and without ETI by immunoimaging, expressed as a percentage of total DAPI+ positive cells. Statistical significance was determined as p-value of < 0.05 using Student’s t test (*: p ≤ 0.05; **: p ≤ 0.01; ***: p ≤ 0.001 C) Microscopy of VeroE6 cells pre-treated with DMSO, CFTR modulator Eleacaftor (VX445, 3μM), Tezacaftor (VX661, 3 μM), Ivacaftor (VX770, 1 μM) or ETI (3μM VX445/3 μM VX661/1 μM VX770) prior to infection with SARS-CoV-2 expressing NeonGreen (MOI = 3). Images were taken 2 dpi. D) Percentage of SARS-CoV-2 N positive cells in CF human bronchial epithelial cells treated with and without ETI by immunoimaging, expressed as a percentage of total DAPI+ positive cells. Statistical significance was determined as p-value of < 0.05 using Student’s t test (*: p ≤ 0.05; **: p ≤ 0.01; ***: p ≤ 0.001 E) Percentage of SARS-CoV-2 N positive cells in Non CF human bronchial epithelial cells treated with and without ETI by immunoimaging, expressed as a percentage of total DAPI+ positive cells. Statistical significance was determined as p-value of < 0.05 using Student’s t test (*: p ≤ 0.05; **: p ≤ 0.01; ***: p ≤ 0.001
