Abstract

A) Representative Immunofluorescence of CF and CFTR-corrected iPSC airway pretreated with vehicle control (DMSO) or CFTR-modulator ETI (3 μM VX445/3 μM VX661/1 μM VX770) and subsequently exposed to Poly(I:C) at 10ug/ml for 1 hour and subsequently stained for p65/RelA (green) and actin (red). Nuclei are stained with DAPI (gray).B) Immunofluorescence analysis of CF and CFTR-corrected iPSC airway treated with vehicle control (DMSO) or CFTR-modulator ETI (3 μM VX445/3 μM VX661/1 μM VX770) stained for Ly6E (green). Nuclei are stained with DAPI (gray). Samples underwent confocal microscopy with z stack imaging of full epithelial layer. Zen Blue (Ziess) software was used to quantify Alex 488 intensity as proportion of image sections. Representative maximum intensity projection images shown Scale bar = 25μm
