Validated HPTLC analysis method for quantification of variability in phytochemicals present in the leaf of Moringa oleifera Lam. Collected from different geographical region of Chhattisgarh, India
Abstract
The leaves of Moringa oleifera, a plant well-known for its health benefits. In order to assess the effects of regional variations on the phytoconstituents found in Moringa oleifera Lam leaves, this work sought to develop a simple, sensitive, and accurate HPTLC method. The following solvent systems were employed to estimate the amounts of the marker medicines rutin and benzyl isothiocyanate in Moringa using the following methodology: Formic acid, methanol, toluene, ethyl acetate, and ethanol (4:3:2:1:0.2) v/v/v/v. Ethyl acetate: chloroform: methanol: formic acid (5:3:2:1) v/v/v/v was the solvent system used for chlorogenic acid, and toluene: ethyl acetate: methanol and formic acid (6:3:1:0.3) v/v/v/v was the solvent system best suited for the separation and measurement of β-sitosterol in leaf extract. Trial runs was held to optimise every solvent system. The Rf value for rutin is 0.21, 0.67 for β-sitosterol, 0.84 for benzoyl isothiocyanate, and 0.34 for chlorogenic acid.
