Abstract

This study used the ligature-induced periodontal injury model to describe cellular and molecular changes during the course of the model. The ligature was placed on the left second upper molar of wildtype mice. Live microCT scanning was performed at days 7, 14, 21. At day 21, the ligature was removed and mice were kept until day 35. Additional live microCT scans were performed at days 28 and 35. Following euthanasia and perfusion, the maxilla was prepared for histological analysis to assess molecular changes. Subsequently, Bmp2 or Bmp7 was deleted in Gli1-positive periodontal progenitor cells using a conditional allele for Bmp2 or Bmp7, Gli1CreERT2 (tamoxifen-induced deletion), and mT/mG lineage tracing. In this experiment, microCT was only performed at days 0 and 35. Changes in bone remodelling and repair were assessed.
