Abstract

A: Multivariate Analysis of Variance (MANOVA) evaluation of all quantified Ca2+ puff parameters (number of Ca2+ puffs, number of Ca2+ puff sites, average Ca2+ puff amplitude and average Ca2+ puff duration) of the vector control conditions of all three HEK3KO (+hIP3R1, +mIP3R2, +hIP3R3) cell lines thereby visualizing group separation. The x- and y- axis show the first and second linear discriminant function from a Linear Discriminant Analysis (lda.LD1 and lda.LD2) respectively. Each data point represents all data quantified from a single cell measurement. (MANOVA, Pillai's trace = 0.51513, p = 9.947e-11). B: Quantification of the number of Ca2+ puffs observed in single HEK3KO (+hIP3R1, +mIP3R2, +hIP3R3) cells transfected with pCMV24-p2a-mCherry (Ctrl). Cell were stimulated through UV-flash photorelease of ci-IP3. Each data point represents a value obtained from one selected, mCherry positive cell. Different point shapes and colors correspond to distinct dates of acquisition (post-hoc one-way ANOVA, N≥28, * shows p
