Abstract
The contraction of slices was measured continuously throughout the 48h of the protocol using the software provided by InVitroSys. Collected data was exported to LabChart (version 8, AD instruments) and analyzed using the Peak Analysis module (MLS380/8, AD instruments). Time to peak, time to 50% relaxation (t50%), and maximum force generated were averaged from ten consecutive contractions extracted from 2, 20, and 40 hours. Tension refers to the total force generated by each slide, normalized for the cross-sectional area of each slide. Bioinformatic statistical approaches are described above. All tests were performed in GraphPad Prism (version 8, Dotmatics) using a Student’s T-test. P values below 0.05 were considered significant. All data is shown as mean± standard error of the mean (SEM). Correlations were measured using a Spearman correlation assay.
