RNA-Seq data from Zourray et al
Abstract
This dataset contains bulk RNA-sequencing data generated for the study “Prenatal sodium channel dysfunction in Dravet syndrome alters cortical development.” RNA-seq was performed on human cortical organoids and cortical organoid assemblies derived from induced pluripotent stem cells (iPSCs) from two individuals with Dravet syndrome and their corresponding isogenic control lines. Libraries were prepared from total/bulk mRNA and sequenced as paired-end reads. The dataset is intended to enable transcriptomic analyses of early cortical developmental changes associated with Dravet syndrome in an isogenic background. The data provided here is the unnormalized count matrix produced with the following workflow: Trimmomatic > STAR aligned to hg38 > FeatureCounts using Gencodev46. STAR was used with the default settings. For feature counts the following setting were used: -B Require both reads in a paired-end fragment to be successfully aligned; -s Library strandness setting (0 = unstranded); -p Indicates paired-end sequencing data; -M Include multi-mapping reads in counting; --fraction; Assign fractional counts to multi-mapping reads (e.g. 1/N per alignment)
