Abstract
Title: Gly-beta-MCA modulates bile acid metabolism to reduce hepatobiliary injury in Mdr2 KO micePMID: 40418643DOI: 10.1152/ajpgi.00044.2025Mice and treatments. The Mdr2 KO mice (Strain #:002539) on congenic FVB/N genetic background were purchased from the Jackson Laboratory (Bar Harbor, ME). Heterogenous mice were bred to generate homozygous Mdr2KO mice and WT littermates. Mice were housed in micro-isolator cages under 7 am - 7 pm light cycle and 7 pm -7 am dark cycle. Gly-bMCA was mixed with chow diet to achieve ~160 mg/kg/day dose, which was calculated based on 4 g/day food intake by a 25 g mouse. Gly-bMCA treatment was initiated when mice were 6 weeks old and lasted for 4 weeks. Tissues and blood samples were collected after mice were fasted for 6 hours from 9 am to 3 pm and euthanized by isoflurane inhalation. Animals received humane care according to the criteria outlined in the “Guide for the Care and Use of Laboratory Animals.” All animal studies were approved by the Institutional Animal Care and Use Committee of the University of Oklahoma Health Sciences Center (Approval # 22-072-EAFHI).
