Abstract

Sample Prep: Dilute serum/plasma 1:101 (e.g., 10μl sample + 1.0ml sample buffer).Incubation: Incubate diluted samples in the microplate for at least 10 minutes at room temperature (+18∘C to +25∘C). Procedure:First Incubation: Incubate diluted samples (100 µl) in the coated wells for 60 min at 37∘C.Wash: Wash 3 times with 300 µl wash buffer.Second Incubation: Add 100 µl Enzyme Conjugate and incubate for 30 min at 37∘C.Wash: Wash 3 times.Substrate Reaction: Add 100 µl TMB substrate and incubate for 15 min at room temperature.Stop: Add 100 µl Stop Solution.Readout: Read absorbance at 450 nm (reference wavelength 620 nm) within 30 min.Storage: The kit must be stored at +2∘C to +8∘C.Limitations: IgM detection is not sufficient for definitive confirmation, and results should be interpreted alongside clinical symptoms. Interpretation of Results as shown in the manufactor instructions handout. Ratio < 0.8: Negative.Ratio 0.8 to < 1.1: Borderline/Equivocal.Ratio ≥ 1.1: Positive.
