LC-HRMS targeted lipidomics of mouse brain after VPS13A knockdown
Abstract
This dataset presents a targeted LC-HRMS lipidomics analysis of mouse brain tissue investigating lipid metabolic alterations associated with neuronal VPS13A deficiency. VPS13A, a lipid transfer protein involved in intermembrane lipid transport, plays a critical role in maintaining membrane organization and neuronal function; its loss is linked to dysfunction in vulnerable neural circuits. In this study, VPS13A knockdown was induced via stereotaxic injection of an AAV8 vector expressing VPS13A-specific shRNAs in neurons, using scrambled shRNA as control. Lipidomic profiling was performed on pooled frontal cortex and striatum samples from 12 mice (6 knockdown and 6 control; balanced by sex), enabling the detection of early changes in lipid composition and homeostasis. A total of 172 lipid species across 17 lipid classes were quantified using class-specific internal standards and normalized to protein content (final units, pmol/mgprot), providing insight into lipid alterations that may contribute to the molecular mechanisms underlying VPS13A-related neuronal dysfunction.
