Abstract

Data visualization for new-EEC relative position (Extended Data Fig. 1a) and clone distribution (Fig. 2j and Extended Fig. 4a,b,c and 6a) was implemented via the R ggplot2 package. Detailed parameters are provided in the scripts 1. New-EEC relative-position distribution.R and 2. Clone_distribution.R. 1. New-EEC relative-position distributionData visualization of the relative positions of new-EECs was performed using the ggplot2 package in R. Datasets included normal conditions (5 d, 10 d, 14 d AE) and post-injury treatments (3 d, 5 d, 10 d AE) from the spatial quantification of new-EECs (RFP⁺ Pros⁺) in ReDDM-traced midguts (Extended Data Fig. 1a). Vertical jitter (mutate(y_position = rnorm(n(), 0, 0.08))) was applied to avoid overlapping data points. 2. Clone distribution1. Clone data from start-point labeled MARCM 19A were visualized (Fig. 2j and Extended Fig. 6a). The x-axis represented days after eclosion and the y-axis represented clone size (Cell_number). Samples were grouped by EEC_ number, with colors #808080, #0000FF, #FF0000 and #00FF00 corresponding to groups of 0, 1, 2 and >2 EECs, respectively. The plots illustrated changes in clone size and intraclonal EEC number over time post eclosion.2. Visualization was performed using clone data from start-point labeled MARCM 82B (nlsGFP) (Extended Fig. 4a,b,c). In Extended Fig. 4a, gut region was set as the x-axis and clone size (Cell_ number) as the y-axis; samples were categorized by ISC_ number and colored with #898989, #8FC31F, #00A0E9 and #EA5514 for 1, 2, 3 and 4 ISCs, showing clone distribution across distinct gut regions. In Extended Fig. 4b, days after eclosion was used as the x-axis with the same grouping and color scheme, displaying the distribution of clone size and Dl⁺ cell number over time post eclosion. In Extended Fig. 4c, ISC_ number was plotted on the x-axis and clone size on the y-axis; samples were grouped by EEC_ number using the aforementioned color palette to show the distribution of clone size and EEC number across different ISC counts. For all plots above, geom_jitter(shape = 1, size = 2, width = 0.3, height = 0.2, alpha = 1) was applied to prevent overlapping data points.
