Abstract

Extended Data associated with "Enhancer-gene regulatory interactions in colorectal cancer revealed through genome-wide CRISPRi perturbations" published in Wellcome Open Research. The 238 significant (FDR < 0.1) results from the SCEPTRE analysis annotated with functional data. "N_nonzero_trt" and "n_nonzero_cntrl" indicate the number of cells that express the gene, with and without the gRNA, respectively. “ABC Score” provides the score from the ABC analysis determining if the enhancer is affecting expression of the gene. The default threshold of 0.025 was used. “Micro-C” indicates if the enhancer region has a 3D contact with the TSS of the target gene. “Intronic” indicates whether the enhancer falls within the intron of the target gene. “Essential gene” indicates if the gene was determined to be essential in the DepMap CRISPR knockdown screens - HT29: essential in the HT29 cell line; Primary only: only primary CRC cell lines; Metastasis only: only in metastatic CRC cell lines; Primary&Metastasis: essential in both primary and metastatic CRC cell lines. The “Fitness score” columns refer to the target priority scores from DepMap, either in the CRC cell lines, or across all cancers (pancancer). A score of 40 is indicative of a potential therapeutic target.
