Abstract

To investigate association of CYP2D6 gene variants rs1065852 with labetalol responsiveness in PEC patients of Khyber Pakhtunkhwa, Pakistan.A prospective cohort study was conducted on 87 PEC Patients. Demographic and Clinical variales were recorded. Genotyping of the CYP2D6 rs1065852 variant was performed and two additional SNPs (rs769258 and rs771774661) were identified in same region. Statistical analysis was performed using SPSS version 25, including logistic regression adjusted for clinical confounders. No significant associations between labetalol response and clinical variables. However, a significant association was observed between the CYP2D6 rs1065852 variant and labetalol response. The G allele was more frequent in non-responders (59.3%) than responders (40.6%) (Adjusted OR = 4.324, 95% CI: 1.556-12.021, p = 0.005). The GG genotype was predominant among non-responders (71.87%) compared to responders (56.52%), whereas the AA genotype was more common in responders (39.13%). The additional SNPs identified, rs769258 and rs771774661, showed consistent patterns of co-occurrence with rs1065852 genotypes across study population. The study provides evidence that presence of A allele in CYP2D6 rs1065852 influences labetalol’s efficacy in controlling SBP and DBP in PEC patients.
