Abstract

Plant-derived small molecules continue to gain prominence as strategic starting points in drug discovery due to their system-level biological activity. These compounds occupy an underexplored chemical space that is highly compatible with complex protein architectures and dynamic cellular networks. Their ability to involve multiple targets simultaneously aligns with emerging therapeutic paradigms that prioritize pathway modulation over single enzyme inhibition for multifactorial diseases. Advances in semi-synthetic chemistry, artificial intelligence–driven molecular design, and high-resolution screening platforms can reshape natural product research into a predictive and optimization-oriented discipline. Innovations in formulation science and systems pharmacology are addressing longstanding translational barriers, enabling more consistent, scalable, and mechanism-based development. These approaches position plant-derived molecules as rational, data-integrated candidates in modern pharmaceutical pipelines rather than empirical interventions.
