Abstract

Background: Incised wounds are open wounds that are at risk of infection and delayed healing. The use of topical formulations derived from natural ingredients has emerged as an alternative therapy to support wound healing. The stem bark of the oil mangrove (Rhizophora apiculata) contains bioactive compounds that may accelerate wound healing, while emulgel was selected as a topical dosage form due to its good stability, ease of application, and ability to enhance the penetration of active compounds. Methods: This study was a laboratory experimental study with a post-test only control group design. A total of 24 male white rats were divided into six groups: normal control, positive control, negative control, and three treatment groups receiving emulgel containing 96% ethanol extract of oil mangrove stem bark at concentrations of 1% (P1), 2.5% (P2), and 5% (P3). Incised wounds were created in a controlled manner and treated for 14 days. Observed parameters included wound healing phases, wound contraction and length reduction, wound healing duration, and macroscopic assessment using the Nagaoka scoring system. Results: The treatment groups showed a faster incised wound healing process compared to the negative control group, as indicated by a more rapid transition of healing phases, greater wound contraction and shortening, and a shorter healing duration. The P3 group demonstrated the shortest mean healing time (8.50 days) compared to the negative control (11.50 days). Statistically significant differences were found in wound contraction and healing duration among groups (p < 0.05), while no significant differences were observed in Nagaoka scores (p > 0.05). Conclusions: Emulgel containing 96% ethanol extract of oil mangrove (Rhizophora apiculata) stem bark is effective in accelerating incised wound healing in male white rats, with the 5% concentration (P3) providing the best results, without significant differences in Nagaoka scores among groups during the observation period.
