Abstract

Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants increasingly linked to metabolic disturbances. This study investigated how microbial metabolites generated during in vitro colonic fermentation under PFAS exposure affect hepatocyte metabolism. Fecal samples from nine different donors of different sexes and dietary backgrounds were fermented with or without PFAS. The resulting extracts were applied to HepaRG hepatocytes for 24 h. Metabolomic and lipidomic profiling revealed distinct alterations compared to PFAS-spiked controls. On the one hand, bile acids, namely cholic acid (log2(FC) = 1.04
