Molecular Docking
Abstract
Molecular docking is a fundamental computational technique in modern drug discovery to predict molecular interactions between ligands and target protein receptors. This method allows researchers to efficiently virtual screen thousands of drug candidate compounds before experimental testing. This article discusses the basic principles of molecular docking, the methodology in Molecular Operating Environment (MOE) software, and crucial validation aspects to ensure the reliability of prediction results. The discussion includes the concept of ligand-receptor binding prediction, the role of docking in virtual screening, ligand and receptor structure preparation, the process of identifying active sites with Site Finder, docking simulations, binding pose determination, and validation methods using redocking and cross-docking. Molecular docking has proven to be a powerful tool in accelerating the drug discovery process by reducing the cost and time required in the development of new therapeutic candidates.
